چکیده

There are two distinct types of adipose tissue which have different functions within the body, white (WAT) and brown (BAT). Browning of WAT occurs with increases in the WAT sympathetic nervous system (SNS) drive. In this regard we previously reported that melatonin (MEL) stimulation of MEL receptor 1A (MEL1a) within the SNS outflow to the WAT might be implicated in a naturally-occurring reversal of obesity (by ~30% of total body fat). Therefore, in this study we tested the hypothesis that MEL causes browning of WAT through the stimulation of SNS drive to WAT. This was done by comparing specific browning and lipolytic markers in WAT following 10 weeks of MEL treatment, short day housing (SD), and long day housing with saline injections (LD+VEH). Browning effects of a 5 day treatment of a β3-adrenergeric (β3AR), CL 316, 243, were also measured. We found that CL 316, 243, MEL treatment, and SD housing had increased expressions of browning markers within WAT and lipolytic activity in MEL treated animals was increased in specific WAT. Wnt inhibition enhances browning of mouse primary white adipocytes

The global epidemic in obesity and metabolic syndrome requires novel approaches to tackle. White adipose tissue, traditionally seen as a passive energy-storage organ, can be induced to take on certain characteristics of brown fat in a process called browning. The "browned" white adipose tissue, or beige fat, is a potential anti-obesity target. Various signaling pathways can enhance browning. Wn...

O-GlcNAc Transferase Enables AgRP Neurons to Suppress Browning of White Fat

Induction of beige cells causes the browning of white fat and improves energy metabolism. However, the central mechanism that controls adipose tissue browning and its physiological relevance are largely unknown. Here, we demonstrate that fasting and chemical-genetic activation of orexigenic AgRP neurons in the hypothalamus suppress the browning of white fat. O-linked β-N-acetylglucosamine (O-Gl...

The browning of white adipose tissue: some burning issues.

Igniting thermogenesis within white adipose tissue (i.e., promoting expression and activity of the uncoupling protein UCP1) has attracted much interest. Numerous "browning agents" have now been described (gene ablations, transgenes, food components, drugs, environments, etc.). The implied action of browning agents is that they increase UCP1 through this heat production, leading to slimming. Her...

The tumor secretory factor ZAG promotes white adipose tissue browning and energy wasting.

Cachexia is a complex tissue-wasting syndrome characterized by inflammation, hypermetabolism, increased energy expenditure, and anorexia. Browning of white adipose tissue (WAT) is one of the significant factors that contribute to energy wasting in cachexia. By utilizing a cell implantation model, we demonstrate here that the lipid mobilizing factor zinc-α2-glycoprotein (ZAG) induces WAT brownin...

Osteogenic Differentiation of Rat Mesenchymal Stem Cells from Adipose Tissue in Comparison with Bone Marrow Mesenchymal Stem Cells: Melatonin As a Differentiation Factor

Background: Adipose-derived stem cells (ADSC) could be an appealing alternative to bone marrow stem cells (BMSC) for engineering cell-based osteoinductive grafts. Meanwhile, prior studies have demonstrated that melatonin can stimulate osteogenic differentiation. Therefore, we assayed and compared the melatonin effect on osteogenic differentiation of BMSC with that of ADSC. Methods: Mesenchymal...
